Cholic acid, also known chemically as 3α,7α,12α-trihydroxy-5β-cholan-24-oic acid, is a primary bile acid naturally synthesized in the liver from cholesterol. It is a white crystalline substance that is insoluble in water but soluble in alcohol and acetic acid. Salts of cholic acid are called cholates. Along with chenodeoxycholic acid, cholic acid is one of the two major bile acids in humans, existing in roughly equal concentrations. It plays a central role in bile acid metabolism, facilitating the digestion and absorption of dietary fats and fat-soluble vitamins.
Cholic acid derivatives are formed from cholyl-CoA, which conjugates with glycine or taurine to produce glycocholic and taurocholic acids, respectively. Functionally, cholic acid regulates bile acid synthesis by downregulating cholesterol-7-α-hydroxylase, the rate-limiting enzyme in bile acid production. This regulation is essential for maintaining cholesterol and bile acid homeostasis in the body. While cholic acid is crucial for normal liver and digestive function, other bile acids, such as chenodeoxycholic acid, are used therapeutically for specific conditions like gallstones due to their ability to modify bile composition.
