Product Description:
Levorphanol, marketed under the brand name Levo-Dromoran, is a potent opioid analgesic used for the management of moderate-to-severe pain, including chronic, post-surgical, and cancer-related pain. It is the levorotatory enantiomer of racemorphan and is approximately eight times more potent than morphine at the μ-opioid receptor (MOR), providing effective pain relief at lower doses.
Levorphanol exerts its analgesic effects primarily through agonism of the μ-opioid receptor, while also acting on δ-opioid (DOR) and κ-opioid (KOR) receptors, as well as the nociceptin receptor (NOP). Additionally, it functions as an NMDA receptor antagonist and a serotonin-norepinephrine reuptake inhibitor (SNRI), which may enhance its analgesic and neuropathic pain-modulating properties. At very high concentrations, levorphanol can also act as a glycine and GABA receptor antagonist.
Administered orally or via injection under strict medical supervision, levorphanol provides extended pain relief due to its long half-life. Its high potency, multimodal mechanism, and efficacy in severe pain management make it a valuable therapeutic option for patients requiring step 3 opioids according to the World Health Organization pain ladder.
